HMMR gene associated with higher risk of breast cancer in women.

Researchers from different laboratories around the world have identified that variations in the HMMR gene may be associated with higher risk of breast cancer in women. The HMMR gene, newly described as a cancer susceptible gene, demonstrates the link between the risk for breast cancer and centrosome dysfunction.

Variation in the HMMR gene facilitates breast cancer development. In a normal population, the risk of breast cancer may increase by 1.5 times in women who carry the HMMR variation and between 2 to 3 times in women younger than 40 years old. The risk of breast cancer is 23% higher in women who inherit one copy of genetic variant and 46% higher in women who inherit two copies.

Researchers led by Stephen Gruber, M.D., Ph.D., Ph.D., M.P.H., along with researchers from Spain, Israel and other investigation centers in the United States, carried out a study in women from an Ashkenazi Jewish population, known for their higher risk of breast cancer among other cultures and similar women without breast cancer.
Researchers turned to Bioinformatics tools that search for existing data and detect genes with the same functional pattern as well-known breast cancer genes like BRCA1 and BRCA2. Interactions between protein-protein and data from different microarrays analysis were examined and finally 160 genes were related to BRCA1; among those, HMMR was the principal candidate.

HMMR gene plays a key role in centrosome function, responsible for chromosome disjunction in cellular division. Alterations in the structure or number of chromosomes represent an important factor in breast cancer. This study indicates that the variability in risk causes the overexpression of the HMMR gene and in this way, promotes errors in the segregation of genetic material during cellular division.

Furthermore, like it frequently occurs with different genetic variability that increases the risk of breast cancer, it has been seen that HMMR variability is more common in young women. In older women, environmental factors contribute to cancer development. This explains that, among the women in the study, the younger ones (<40 years old) will show almost two times the risk of developing breast cancer than older women. However, this data corresponds to a Jewish population and it should be analyzed in different populations and further investigated if their impact is higher or lower than in the Jewish population.

The influence of HMMR variability in breast cancer forecast is one of the most important aspects that are unknown and which should be analyzed in more detail. If more studies are done under this subject, the probabilities to find new treatments for breast cancer will unquestionably increase.

Breast cancer has been an important topic in my life, since it was discovered in my aunt a year ago. In my opinion, this study should be performed in more detail to discover more about the interaction between the HMMR gene and cancer genes that can be a hotspot for the detection and treatment of cancer. Further studies should be executed in some other tissues, such as colon and skin, as this new gene may be related to other types of cancer.
